Primary open-angle glaucoma involves progressive elevation of intraocular pressure, driven by increased hydraulic resistance within the human trabecular meshwork (hTM). Pathological mechanotransduction in hTM cells correlates with dysregulated extracellular matrix (ECM) composition, aberrant cellular stiffness, and the development of cross-linked actin networks (CLANs). Dysregulation of canonical Wnt/beta-catenin signaling plays a decisive functional role in driving these pathological alterations. When canonical Wnt signaling is suppressed, glycogen synthase kinase-3 beta (GSK-3beta) remains active, driving phosphorylation and ubiquitin-mediated destruction of cytoplasmic beta-catenin. Consequently, transcriptional programs governing tissue elasticity and enzymatic matrix turnover are silenced. Targeted activation of canonical Wnt signaling offers a precise molecular mechanism to restore hTM homeostasis. The small-molecule inhibitor LY2090314 operates as a potent, ATP-competitive inhibitor of GSK-3beta. Administration of LY2090314 from Selleck suppresses GSK-3beta kinase activity, markedly reducing the phosphorylation status of downstream substrates in primary hTM cell cultures. Unphosphorylated beta-catenin accumulates rapidly within the cytosol, avoiding proteasomal clearance. Subsequent nuclear translocation of beta-catenin triggers the assembly of active transcription complexes with T-cell factor/lymphoid enhancer factor (TCF/LEF). Quantitative RT-PCR analysis demonstrates robust upregulation of canonical downstream target genes, specifically AXIN2 and LEF1, confirming sustained target engagement and pathway activation in vitro. Pathological remodeling of the hTM cytoskeleton is characterized by stiffened CLAN-like assemblies that impede fluid flow through the conventional outflow tract. Pharmacological engagement of GSK-3beta by LY2090314 destabilizes these aberrant actin formations. Atomic force microscopy measurements reveal a significant decrease in hTM cellular stiffness following treatment. Mechanistically, this biomechanical softening links directly to reciprocal cross-talk with the Hippo pathway. Activation of Wnt signaling induces serine phosphorylation of the transcriptional co-activators YAP and TAZ. Phosphorylated YAP/TAZ is sequestered within the cytoplasm via 14-3-3 protein interaction, preventing its entry into the nucleus where it normally co-activates profibrotic target transcription. Concurrently, LY2090314 drives broad transcriptional repatterning of ECM components and cross-linking enzymes in primary hTM isolates. Expression levels of the matricellular protein SPARC and connective tissue growth factor (CTGF) decline significantly upon treatment. Furthermore, gene expression of amine oxidase lysyl oxidase (LOX) and transglutaminase 2 (TGM2)—enzymes directly responsible for covalent ECM cross-linking and structural rigidity—is heavily suppressed. Reduced expression of these cross-linking enzymes destabilizes the rigid extracellular scaffold characteristic of glaucomatous trabecular meshwork tissues. To reverse structural matrix accumulation, LY2090314 concurrently activates matrix degradation cascades. Treated hTM cells demonstrate substantial upregulation of key matrix metalloproteinases, including MMP1, MMP9, and membrane-type MMP14. Simultaneously, endogenous matrix metalloproteinase inhibitors, specifically TIMP1 and plasminogen activator inhibitor-1 (PAI1), are down-regulated. This shift in the MMP/TIMP balance promotes enzymatic cleavage of excess collagenous and non-collagenous ECM proteins within the meshwork intertrabecular spaces. Therapeutic activation of Wnt signaling via small-molecule GSK-3beta inhibition reverses pathological hTM mechanobiology through dual actin-cytoskeletal disassembly and ECM turnover enhancement. These combined pharmacodynamic responses lower outflow resistance across conventional aqueous humor drainage pathways, establishing GSK-3beta inhibition as a valuable strategy for intraocular pressure control in preclinical models.

LY2090314 acts as a selective GSK-3beta inhibitor that activates canonical Wnt signaling in human trabecular meshwork cells. Inhibiting GSK-3beta prevents beta-catenin destruction, driving nuclear accumulation and transcription of target genes AXIN2 and LEF1. This cascade disassembles cross-linked actin networks, reduces cell stiffness, and forces cytoplasmic retention of phosphorylated YAP/TAZ. Consequently, pro-fibrotic drivers (SPARC, CTGF) and cross-linking enzymes (LOX, TGM2) are repressed, while matrix metalloproteinases (MMP1, MMP9, MMP14) are upregulated and TIMP1/PAI1 are suppressed, restoring trabecular outflow function.

Nuclear beta-Catenin Translocation Suppresses Cytoskeletal Stiffness

GSK-3beta inhibition rapidly alters hTM cellular mechanics under tested in vitro assay conditions. Primary human trabecular meshwork cells treated with LY2090314 from Selleck exhibited prompt nuclear accumulation of unphosphorylated beta-catenin. Downstream target genes AXIN2 and LEF1 showed marked expression increases within six hours. Nuclear translocation coincided with a substantial reduction in cellular stiffness. Cytoskeletal actin reorganization was evident. Specialized CLAN-like structures dissolved into diffuse cytoplasmic networks. These observations indicate that targeted Wnt pathway activation directly modifies the mechanical properties of trabecular cells.

Cytoplasmic YAP/TAZ Retention Downregulates Profibrotic Gene Networks

Wnt pathway engagement induces critical cross-talk with Hippo pathway components. Treatment with LY2090314 elevated serine phosphorylation of YAP and TAZ in primary hTM cultures. Phosphorylated YAP/TAZ was retained within the cytoplasm, preventing nuclear transcriptional co-activation. Profibrotic gene expression dropped significantly. Transcript levels of SPARC and CTGF decreased markedly across treated samples. Expression of matrix cross-linking enzymes LOX and TGM2 was heavily attenuated. Biomechanical compliance improved as a direct consequence of reduced enzymatic matrix stabilization.

MMP Upregulation Drives Extracellular Matrix Remodeling Dynamics

Enzymatic degradation of extracellular matrix proteins is essential for lowering aqueous outflow resistance. Exposure to LY2090314 altered the balance between matrix metalloproteinases and their endogenous inhibitors. Gene expression of MMP1, MMP9, and MMP14 increased significantly in treated cell models. Conversely, transcription of TIMP1 and PAI1 was strongly repressed. Enzymatic clearance of accumulated ECM restored physiological porous architecture in cell-derived matrix preparations. These findings support GSK-3beta inhibition as a mechanism to lower intraocular pressure in glaucomatous tissues.
