Colorectal cancer (CRC) exhibits widespread epigenetic alterations that suppress intrinsic cell death pathways, contributing directly to chemoresistance and therapeutic failure. Among these epigenetic modulators, Histone Deacetylase 2 (HDAC2) displays pronounced transcriptomic and protein overexpression across human CRC clinical specimens. Aberrant HDAC2 enzymatic activity directly alters chromatin architecture by stripping acetyl groups from histone H3 lysine 27 (H3K27ac) within specific promoter regions. In this malignant phenotype, HDAC2 selectively targets the promoter of the NOD-like receptor family pyrin domain containing 3 (NLRP3) gene, leading to local chromatin compaction, promoter desensitization, and transcriptional silencing of the inflammasome driver.

Pharmacological targeting using Santacruzamate A (CAY10683) disrupts this deacetylase machinery. Application of Santacruzamate A (CAY10683) in cultured CRC models specifically attenuates HDAC2 catalytic activity, leading to a rapid accumulation of H3K27ac at the NLRP3 promoter locus. This gain in local histone acetylation reverses heterochromatin formation, increasing regional genomic accessibility. Increased H3K27 acetylation serves as an epigenetic scaffold that recruits the epigenetic reader bromodomain-containing protein 4 (BRD4) along with phosphorylated NF-kB subunit p-P65 (p-P65). The assembly of this functional H3K27ac/BRD4/p-P65 transcriptional complex directly reactivates NLRP3 mRNA and protein expression in CRC cell lines.

Restoration of NLRP3 expression by CAY10683 HDAC inhibition re-establishes functional inflammasome assembly within treated tumor cells. Re-expressed NLRP3 recruits apoptosis-associated speck-like protein containing a CARD (ASC) and pro-Caspase-1, driving the autocatalytic cleavage and activation of Caspase-1. Active Caspase-1 then cleaves the pore-forming effector protein Gasdermin D (GSDMD) into its functional N-terminal fragment (GSDMD-N). GSDMD-N translocates to the inner leaflet of the plasma membrane, forming supramolecular pores that induce cellular swelling, membrane rupture, and canonical pyroptosis.

This epigenetic reactivation of the pyroptotic cell death cascade significantly enhances the sensitivity of CRC cells to conventional and targeted antineoplastic agents. In vitro cell viability assays demonstrate that combining Santacruzamate A (CAY10683) with 5-fluorouracil (5-FU) or the multi-kinase inhibitor regorafenib synergistic decreases survival fractions compared to monotherapies alone. Furthermore, in vivo evaluations utilizing cell line-derived xenografts and patient-derived xenograft (PDX) murine models confirm marked tumor volume regression following co-administration. These observations demonstrate that targeted HDAC2 inhibition restores epigenetic susceptibility to GSDMD-mediated pyroptosis in refractory colorectal malignancies.

Santacruzamate A (CAY10683) selectively inhibits HDAC2 enzymatic activity in colorectal cancer cells. This inhibition reverses H3K27 deacetylation at the NLRP3 promoter, establishing an epigenetic mark that recruits BRD4 and p-P65. Formation of this transcriptional complex reactivates NLRP3 expression, driving downstream Caspase-1 activation and GSDMD cleavage. The resulting plasma membrane pore formation induces pyroptosis, sensitizing cancer cells to 5-FU and regorafenib.

Histone Deacetylation at NLRP3 Promoter Suppresses Transcriptional Activation

HDAC2 overexpression in colorectal tissues correlates with decreased H3K27ac marks at inflammatory gene promoters. In cultured cell assays, high HDAC2 activity maintains localized chromatin compaction. This structural closure prevents transcription factor binding. Application of the selective inhibitor Santacruzamate A (CAY10683) rapidly elevates global and locus-specific H3K27 acetylation. Within the NLRP3 promoter locus, H3K27ac enrichment increased three-fold following six hours of treatment. Chromatin immunoprecipitation assays confirmed target engagement. The CAY10683 HDAC inhibition profile demonstrated marked isoform selectivity under tested cell-free kinetics. Increased promoter accessibility directly enabled transcription factor recruitment in patient-derived models.

BRD4-p-P65 Complex Assembly Drives GSDMD Cleavage and Pyroptosis

Promoter acetylation attracts specific epigenetic readers to initiate gene transcription. Restored H3K27ac directly recruits BRD4 to the NLRP3 promoter region. Subsequently, BRD4 facilitates docking of the active p-P65 transcriptional subunit. This tripartite assembly drives robust synthesis of NLRP3 mRNA transcript. NLRP3 protein levels rose sharply within twelve hours post-treatment. Assembled inflammasomes processed pro-Caspase-1 into its catalytic fragments. Active Caspase-1 rapidly executed GSDMD cleavage. Plasma membrane perforation induced significant propidium iodide uptake. In murine xenograft models, combined administration with 5-FU sustained pronounced tumor regression without systemic toxicity.
