High-throughput chemical screening remains a primary strategy for identifying target-directed agents in pediatric neuro-oncology. In a systematic study evaluating 12,800 small molecules, researchers identified candidate therapies against medulloblastoma. Among the most potent hits were two histone deacetylase inhibitors, quisinostat and dacinostat. Utilizing medulloblastoma therapeutic screening studies, investigators validated these compounds in established cell lines, including Daoy and D283. Application of Quisinostat (JNJ-26481585) Dihydrochloride from Selleck reduced cellular viability across tested concentrations in vitro.

Histone deacetylases (HDACs) serve as core epigenetic operators. These enzymes catalyze the removal of acetyl moieties from histone tails, maintaining condensed chromatin that restricts gene transcription. Exposure to Quisinostat (JNJ-26481585) Dihydrochloride reversed this repressive state. Immunoblotting revealed elevated levels of acetylated histone H3 and H4 within hours of target engagement.

Downstream effectors exhibited marked changes following HDAC blockade. Expression of the key oncogenic driver c-Myc was significantly reduced. Simultaneously, phosphorylation of Akt at regulatory sites diminished, interrupting central pro-survival pathways. As a consequence, medulloblastoma cells accumulated in the G2/M phase of the cell cycle, halting cellular proliferation.

Prolonged cycle arrest triggered apoptotic cascades. Enzymatic assays demonstrated activation of caspase-3 and caspase-7. Cleavage of poly(ADP-ribose) polymerase (PARP) confirmed execution of caspase-dependent cell death. In Daoy mouse xenograft models, systemic treatment suppressed tumor growth kinetics. Tumor tissue analyses showed decreased Ki-67 staining and increased cleaved PARP, while animal body weights remained stable throughout the study period.

Quisinostat (JNJ-26481585) Dihydrochloride acts as a pan-HDAC inhibitor. By inhibiting histone deacetylases, it increases histone H3 and H4 acetylation, relaxing chromatin architecture. This epigenetic modification suppresses c-Myc expression and inhibits Akt phosphorylation, blocking survival cascades. Consequently, medulloblastoma cells undergo G2/M cell cycle arrest and caspase-dependent apoptosis.

Histone H3 and H4 Acetylation Attenuates Oncogenic Signaling

Epigenetic dysregulation frequently supports tumor cell survival in pediatric brain tumors. Pharmacological inhibition of histone deacetylases directly modifies transcriptional output. In Daoy and D283 medulloblastoma cell lines, treatment with Quisinostat (JNJ-26481585) Dihydrochloride from Selleck increased core histone H3 and H4 acetylation. Chromatin structure opened following histone hyperacetylation. This epigenetic modification impaired key oncogenic pathways. Protein levels of c-Myc dropped noticeably within hours. Concurrently, signaling cross-talk led to reduced Akt phosphorylation, attenuating downstream kinase activity.

Cell Cycle Arrest and Caspase Activation Drive Tumor Clearance

Interruption of transcriptional activity alters cell cycle dynamics. Flow cytometry revealed that medulloblastoma cells exposed to pan-HDAC inhibitors accumulated predominantly in the G2/M phase. Mitotic entry stopped. Extended cycle arrest initiated programmed cell death pathways. Enzymatic cleavage of PARP confirmed caspase-dependent apoptosis. In Daoy murine xenograft models, daily administration reduced overall tumor volume. Tissue immunohistochemistry confirmed these cellular observations. Ki-67 expression decreased in tumor sections, while cleaved PARP markers increased without altering mouse body weight.
